Laboratoř strukturní chemie

Laboratoř strukturní chemie se zabývá strukturními studiemi membránových a ve vodě rozpustných makromolekulárních komplexů především s použitím metod rentgenové difrakce. Pomocí této metody se dají určit struktury proteinů na atomárním rozlišení. Poznání struktur proteinů je klíčové při zjišťování a popisu detailního mechanismu biologických procesů nebo vývoji terapeutik.


Popis laboratoře:

Laboratoř je rozdělena na tři funkční celky a jednu školící část:

MolBiol - část molekulární biologie, ve které se izolují a purifikují zkoumané proteiny,

 XtallExp - část proteinové krystalizace a krystalografie, kde se provádí krystalizace proteinů s cílem připravit difraktující monokrystaly, které jsou následně měřeny na zdrojích synchrotronového záření a difrakční data softwarově zpracována,

 MolStruct – část molekulárního modelování a strukturních studií, ve které se provádějí následné molekulárně dynamické a strukturní studie.

 XtallTraining - laboratoř dále nabízí ostatním pracovníkům, kteří mají zájem o krystalizaci vlastních proteinů, možnost jejich krystalizace buď přímo našimi zkušenými spolupracovníky, nebo samostatně pod jejich vedením s možností kompletního vyřešení struktury v naší laboratoři.

  • základní výzkum v oblasti izolace a purifikace proteinů a jejich krystalizace
  • řešení struktur z difrakčních dat a strukturní studie zkoumaných proteinů
  • vývoj a testování alternativních a pokročilých krystalizačních technik
  • úzká spolupráce s partnerskými laboratořemi v ČR i zahraničí
  • zaškolení a poskytování servisních služeb v rámci krystalizačních studií proteinů
  • výuka předmětů Anorganická chemie 1 (UCH/100), Anorganická chemie 2 (UCH/101), Organická chemie pro biology (UCH/032) a Biokrystalizační metody (UCH/253)
  • organizace mezinárodního kurzu pod záštitou Federace Evropských Biochemických Společností: FEBS-INSTRUCT practical crystallization course „Advanced methods in macromolecular crystallization“
